10X Tris-CAPS Transfer Buffer (With and Without SDS) for Discontinuous Semi-Dry Blotting (pH 9.6)

DESCRIPTION

The 10X Tris-CAPS Buffer is formulated for Western transfers using a discontinuous system in semi-dry blotting apparatus.

In a semi-dry transfer, the buffer reservoirs are the filter paper on either side of the gel, which are independent (discontinuous). In a discontinuous system, methanol (15%) is included in the buffer on the membrane (anode) side of the blot assembly and SDS (0.1%) is used on the gel (cathode) side, taking advantage of the positive effects of each buffer component.

The discontinuous buffer system increases protein transfer efficiency. In addition, transfers are finalized in a brief period (15–60 min.).

Meb Biosciences 10X Tris-CAPS transfer buffer (without SDS, Cat. No. TSCS01-10X) contains 0.6 M Tris base, and 0.4 M CAPS (pH 9.6 ± 0.1). The buffer can be used for making the anode and the cathode buffers. Prior to use, methanol is added to a final concentration of 15% to the anode buffer (by diluting the buffer 10-folds with deionized water and methanol), and SDS is added to a final concentration of 0.1% in the 1X diluted cathode buffer. Med Biosciences 10X Tris-CAPS transfer buffer (with SDS, Cat. No. TSCS02-10X) contains 0.6 M Tris base, 0.4 CAPS, and 1% SDS (pH 9.6 ± 0.1). This solution can be used for the cathode buffer after 10-fold dilution with deionized water.

We also provide both 10X Tris-CAPS solutions (with and without SDS) together in a kit format containing 500 mL of 10X Tris-CAPS without SDS, and 500 mL of 10X Tris-CAPS with 1% SDS for making the anode and cathode transfer buffers (Cat. No. TSCS03-10X).

Our buffers are made from Highly Pure Molecular-Grade Chemicals in Autoclaved 18.2 mega Ω-cm Water.

The buffers are sterile filtered through 0.22µm membrane.

The buffers are evaluated and certified for the absence of DNase, RNase, and protease activities.
