DESCRIPTION
Phenol red (also known as phenolsulfonphthalein or PSP) is used as a pH indicator. The sodium salt form of phenol red is used for preparation of cell culture media as a visual indicator to monitor shifts from neutral to acidic pH values. It is typically used in cell culture media where ideally a solution of phenol red will have a yellow to orange color at a pH ≤ 6.8, and a gradual increase in red color at pH 7.0 to reach dark red/purple color at pH ≥ 8.2 (please see the chart photograph for reference colors).
Med Biosciences 0.5% Phenol Red Solution is ready-to-use for Routine cell culture work and can be easily diluted to the desired lower concentrations.
Our Phenol Red solutions is prepared from a highly pure reagent with Autoclaved 18.2 mega Ω-cm DNase/RNase-free Water to ensure utmost quality.
The solution is manufactured according to strict quality control standards to ensure lot-to-lot consistency. Our Phenol Red is Certified Sterile, DNase & RNase Free, Protease Free and Endotoxins Free (<0.0050EU/ml) solution. Each lot of our Cell Culture Grade 0.5% Phenol Red Solution is subjected to rigorous quality control validation which involves checking for Endotoxin lipopolysaccharides (LPL), RNase activity, Exonuclease activity and Nonspecific Endonuclease activity, Nonspecific Protease activity and Sterility as shown in the following table.
| Test | Sample Incubated with Phenol Red Solution | Analysis After Incubation | Validation |
|---|---|---|---|
| Endotoxin contamination | Limulus amebocyte lysate | Photometric LAL quantitative test | Pass/Endotoxin Level <0.0050EU/ml |
| RNase activity | Labeled RNA | Electrophoresis (PAGE Analysis) | Pass/No Degradation |
| Exonuclease activity | Linear Double Stranded DNA | Electrophoresis (PAGE Analysis) | Pass/ No Degradation |
| Nonspecific Endonuclease Activity | Supercoiled plasmid DNA | Agarose Gel Electrophoresis | Pass/No Decrease Supercoiling |
| Nonspecific Protease Activity | Purified casein solution | Spectrophotometrically for liberated Tyrosine (tyr) residues from digested casein using Folin-Ciocalteus Phenol reagent (FC) | Pass/No Significant Blue Chromogen formation between Tyr and FC reagent. |
| Sterility | Microbial Growth after plating Phenol Red solution on specific microbiology agar plates (trypticase Soy Agar, and Nutrient Agar) and incubating for 48 hours at 37 á´¼C | Counting colony forming units on cultured plates (CFU) | Negative/No Colony Formation |

NOTES
Important, phenol red in tissue culture media can function as a weak estrogen, a property associated with the presence of lipophilic impurities, especially observed in human breast cancer cells.
Do not autoclave the solution.
Each batch of Phenol Red solution undergoes strict quality control and standard operating procedures to ensure the high quality of the product, and lot-to-lot consistency.
