DESCRIPTION
Tris is a widely used buffer in many research applications of different fields including DNA and RNA molecular biology work, proteins and many other macromolecules purifications and characterization, enzyme kinetics and many immunological assays and FISH protocols.
Med Biosciences 1.0 M Tris buffer meets the research needs for all applications due to the following features:
1. The buffer is provided at different pH from pH 6.5 to pH 9.5.
2. We use ultrapure molecular biology grade Tris-HCl and Tris-Base, and the 1.0 M solution is prepared in Autoclaved 18.2 mega Ω-cm DNase/RNase-free Water to ensure utmost quality. Therefore, our buffer is suitable for all molecular biology applications.
3. Our buffer is sterile, filtered through 0.22 µm filter into a final sterile and DNase/RNase-free bottle. Therefore, there is no need to sterilize by filtration when you dilute the buffer using sterile water.
4. Each lot of our Molecular Biology Grade Tris buffer is subjected to rigorous quality control which involves checking for RNase activity, Exonuclease activity, Nonspecific Endonuclease activity, Nonspecific Protease activity and Sterility as shown in the following table.
Test | Sample Incubated with Tris-HCl or Tris-Base Solution | Analysis After Incubation | Validation |
---|---|---|---|
RNase activity | Labeled RNA | Electrophoresis (PAGE Analysis) | Pass/No Degradation |
Exonuclease activity | Linear Double Stranded DNA | Electrophoresis (PAGE Analysis) | Pass/ No Degradation |
Nonspecific Endonuclease Activity | Supercoiled plasmid DNA | Agarose Gel Electrophoresis | Pass/No Decrease Supercoiling |
Nonspecific Protease Activity | Purified casein solution | Spectrophotometrically for liberated Tyrosine (tyr) residues from digested casein using Folin-Ciocalteus Phenol reagent (FC) | Pass/No Significant Blue Chromogen formation between Tyr and FC reagent. |
Sterility | Microbial Growth after plating Tris solution on specific microbiology agar plates (trypticase Soy Agar) and incubating for 48 hours at 37 á´¼C | Counting colony forming units on cultured plates (CFU) | Negative/No Colony Formation |

NOTES
The pH values of all buffers are temperature and concentration dependent. For Tris buffers, pH increases about 0.03 unit per 1 á´¼C decrease in temperature and decreases 0.03-0.05 unit per ten-fold dilution. For precise applications, use a carefully calibrated pH meter with a glass/calomel combination electrode.
Each batch of the Tris buffer solution undergoes strict quality control and standard operating procedures to ensure high quality and sterility.
