DESCRIPTION
HEPES has been described as one of the best all-purpose buffers available for biological research. HEPES belongs to the zwitterionic Good buffer group and is effective as a buffer at pH 6.8 to 8.2 (pKa 7.55).
HEPES is ideal for cell culturing because of its usefulness in controlling pH changes in presence of carbon dioxide production from cellular respiration. It is also used for freezing protein solutions. The increase in acidity because of freezing observed in many buffers is negligible in the presence of HEPES buffer. HEPES protects enzymes and coenzymes from denaturation and inactivation during freeze-drying. HEPES buffer has a large list of applications in Molecular Biology, Cell Biology, and Immunology research.
Med Biosciences 1.0 M HEPES buffer meets the research needs in most applications:
1. The buffer is provided in different pH from pH 7.0 to pH 8.0 (pH 7.0, 7.2, 7.4, 7.5, 7.6, 7.8 & 8.0).
2. We use ultrapure molecular biology grade HEPES, and the 1.0 M solution is prepared in Autoclaved 18.2 mega Ω-cm DNase/RNase-free Water to ensure utmost quality. Therefore, our buffer is suitable for all molecular biology applications.
3. Our buffer is sterile, filtered through 0.22 µm filter into a final sterile and DNase/RNase-free bottle. Therefore, there is no need to sterilize by filtration when you dilute the buffer using sterile water.
4. Our HEPES buffer is DNase/RNase and Protease free. The buffer is also Endotoxin-free (Endotoxin Level <0.0050EU/ml, determined by using Limulus amebocyte lysate). Therefore, our buffer is suitable for cell culture applications, DNA and RNA molecular analysis, biochemical and protein purifications and storage, cell imaging, staining and flow cytometry studies, and various immunological applications. 5. Each lot of our Molecular and Cell Culture Grade 1.0 M HEPES Solution is subjected to rigorous quality control which involves checking for Endotoxin lipopolysaccharides (LPL), RNase activity, Exonuclease activity and Nonspecific Endonuclease activity, Nonspecific Protease activity and Sterility as shown in the following table.
Test | Sample Incubated with HEPES Solution | Analysis After Incubation | Validation |
---|---|---|---|
Endotoxin contamination | Limulus amebocyte lysate | Photometric LAL quantitative test | Endotoxin Level <0.0050EU/ml |
RNase activity | Labeled RNA | Electrophoresis (PAGE Analysis) | Pass/No Degradation |
Exonuclease activity | Linear Double Stranded DNA | Electrophoresis (PAGE Analysis) | Pass/ No Degradation |
Nonspecific Endonuclease Activity | Supercoiled plasmid DNA | Agarose Gel Electrophoresis | Pass/No Decrease Supercoiling |
Nonspecific Protease Activity | Purified casein solution | Spectrophotometrically for liberated Tyrosine (tyr) residues from digested casein using Folin-Ciocalteus Phenol reagent (FC) | Pass/No Significant Blue Chromogen formation between Tyr and FC reagent. |
Sterility | Microbial Growth after plating HEPES solution on specific microbiology agar plates (trypticase Soy Agar) and incubating for 48 hours at 37 OC | Counting colony forming units on cultured plates (CFU) | Negative/No Colony Formation |
