DESCRIPTION
The 10X STE buffer solution (Saline-Tris-EDTA Buffer Solution) is a commonly used buffer in molecular biology research. STE is also known as a TEN (Tris-EDTA-NaCl) buffer.
STE buffer is used for DNA extraction protocols. It is used in genomic DNA extraction protocols. It is utilized as a lysis buffer for eukaryotic cells and in the alkaline lysis protocol for M13 isolation.
The buffer is used in genomic DNA extraction because it provides the appropriate pH and salt concentration to effectively solubilize and purify DNA from cells. The Tris component helps to maintain a neutral pH, the EDTA chelates divalent cations that can interfere with the extraction process, and the NaCl provides an appropriate salt concentration for the DNA to remain in solution.
The buffer is also used with Qiagen plasmid kits for resuspension and storage of isolated DNA.
The 10X buffer is composed of 100 mM Tris-Base, 1 M NaCl, and 10 mM EDTA (pH 8.0 ± 0.1).
To use, dilute 1 volume of the 10X STE with 9 volumes molecular biology grade water. The final concentrations in 1X solution are: 10 mM Tris-Base, 100 mM NaCl, and 1 mM EDTA (pH 8.0 ± 0.1).
Our buffer is formulated from Highly Pure Molecular-Grade Chemicals in Autoclaved 18.2 mega Ω-cm Water.
The buffer is sterile, filtered through 1 µm membrane and autoclaved.
Our 10X STE buffer is tested and certified for the absence of RNase/DNase and protease activities.
