DESCRIPTION
Imidazole is used as a buffer in the pH range of 6.2-7.8 at 25 OC. It is recommended as a buffer component for the assay of horseradish peroxidase and as a chelator for the binding of various divalent cations. Imidazole aids the elution of histidine-containing proteins from divalent cation resins (Nickel affinity gel) and can also be used in reverse staining of SDS-PAGE gels for the detection of proteins.
Med Biosciences 2.0 M Imidazole Buffer Solution is provided at different pH from pH 6.5 to pH 7.9.
We use ultrapure molecular biology grade imidazole, and the 2.0 M solution is prepared with Autoclaved 18.2 mega Ω-cm DNase/RNase-free Water to ensure utmost quality. Our buffer is sterile, filtered through 0.22 µm filter into a final sterile and DNase/RNase-free bottle. The buffer is certified being DNase/RNase-free, Protease free, and phosphatase free. Therefore, our buffer is suitable for most molecular biology applications.
Each lot of our Molecular Biology Grade Imidazole buffer is subjected to rigorous quality control which involves checking for RNase activity, Exonuclease activity, Nonspecific Endonuclease activity, Nonspecific Protease activity, phosphatase activity, and Sterility as shown in the following table.
Test | Sample Incubated with Imidazole Solution | Analysis After Incubation | Validation |
---|---|---|---|
RNase activity | Labeled RNA | Electrophoresis (PAGE Analysis) | Pass/No Degradation |
Exonuclease activity Nonspecific Endonuclease Activity |
Linear Double Stranded DNA Supercoiled plasmid DNA |
Electrophoresis (PAGE Analysis) Agarose Gel Electrophoresis |
Pass/ No Degradation Pass/No Decrease Supercoiling |
Nonspecific Protease Activity | Purified casein solution | Spectrophotometrically for liberated Tyrosine (tyr) residues from digested casein using Folin-Ciocalteus Phenol reagent (FC) | Pass/No Significant Blue Chromogen formation between Tyr and FC reagent. |
Nonspecific Phosphatase | p-nitrophenyl phosphate | Spectrophotometrically for the generated p-nitrophenol after removing the phosphate group by the potential phosphatases. | Pass/No Significant yellow Chromogen formation due to liberation of p-nitrophenol |
Sterility | Microbial Growth after plating imidazole solution on specific microbiology agar plates, such as trypticase Soy Agar and nutrient Agar and incubating for 48 hours at 37 ᴼC | Counting colony forming units on cultured plates (CFU) | Negative/No Colony Formation |

NOTES
The pH values of all buffers are temperature and concentration dependent. For Imidazole, pH increases about 0.02 unit per 1 ᴼC decrease in temperature and decreases 0.02-0.04 unit per ten-fold dilution. For precise applications, use a carefully calibrated pH meter with a glass/calomel combination electrode.
Each batch of the Imidazole buffer solution undergoes strict quality control and standard operating procedures to ensure high quality and sterility.
