DESCRIPTION
2X LDS Sample Buffer is used to prepare protein samples for denaturing gel electrophoresis with Bis-Tris or Tris-Acetate gels.
The sample buffer contains lithium dodecyl sulfate (LDS), pH 8.4, which allows for maximum activity of the reducing agent.
The 2X LDS Sample Buffer contains 494 mM Tris, 102 mM EDTA,4% LDS (Lithium dodecyl sulfate), 20% Glycerol, 0.44% Coomassie Brilliant Blue G250, and 0.35 mM Phenol red (pH 8.4 ± 0.1). The sample buffer does not contain any reducing agent.
The final concentrations in 1X LDS Sample buffer are: 247 mM Tris, 0.51 mM EDTA, 2% LDS (Lithium dodecyl sulfate), 10% Glycerol, 0.22% Coomassie Brilliant Blue G250, and 0.175 mM Phenol red (pH 8.4 ± 0.1).
Coomassie Brilliant Blue G250 gives a sharp dye front with both MES-SDS and MOPS-SDS running buffers and migrates much closer to the moving ion front than bromophenol blue. Bromophenol blue runs more slowly than some peptides with MES-SDS Running Buffer. This ensures that small peptides do not run off the gels.
Our buffer is formulated from Highly Pure Molecular-Grade Chemicals in Autoclaved 18.2 mega Ω-cm Water.
The sample buffer is tested and certified for the absence of RNase/DNase and protease activities.
PROCEDURE:
Dilute 1 volume of the 2X LDS Sample Buffer with 3 volumes of sample solution (Example: add 10 µL of the 2X sample buffer to each 10 µL sample).
For reducing conditions, add reducing agents such DTT, TCEP [at final concentration of 50 mM (sold separately at a concentration of 500 mM)] or Beta mercaptoethanol (2%) to reduce disulfide bonds in protein samples. The high pH 8.4 of this buffer enhances the activity of the reducing agent.
For dry or precipitated protein samples, dilute the sample buffer to 1X using ultrapure deionized water and the proper concentration of the reducing agent. Then dissolve the protein sample in an appropriate amount of the diluted sample buffer.
Samples are heated in the 1X LDS Sample buffer for 10 minutes at 70 ᴼC to aid in the denaturation. At this point, samples can remain at room temperature if they are to be used immediately or maybe stored at 4 ᴼC or –20 ᴼC for later analysis.
