SPECIFICATION
Tricine SDS Sample Buffer (with reducing agent) is used to prepare protein samples for denaturing gel electrophoresis using Tris-Tricine denaturing PAGE for separating peptides and small size proteins (˂40kD).
The Tricine sample buffer is formulated based on the method of Schagger von Jagow (1987) with special modifications in our laboratory to grant optimal band resolution and bands tightness when preparing peptides and small proteins for denaturing PAGE with Tris-Tricine-SDS running buffer (please see our running buffer, products Cat. No. BTTS70-10X and Cat. No. BTTS70-1X).
Tricine-SDS sample buffer is compatible to prepare samples for loading on both home-made and precast Tris-Tricine gels (ready-made) such as Mini-PROTEAN® and midi Criterion™ gels from BioRad, Invitrogen Novex Tricine Gel from ThermoFisher Scientific, TEO Tricine Precast Gels from Abcam, and other commercially available Tris-Tricine precast gels.
The 2X Reducing Tricine-SDS sample buffer is composed of 200 mM Tris-HCl, 40% glycerol, 2% SDS, 0.04% Coomassie Blue G-250, and 2% (284 mM) β-mercaptoethanol [Cat. No. BTSSB71-2X-(5X1.5ML, 10ML, 25ML, or 50ML)], or 125 mM Dithiothreitol [DTT, Cat.No. BTSSB72-2X-(5X1.5ML, 10ML, or 25 mL)], pH 6.8. Our sample buffer is formulated from Highly Pure Molecular-Grade Chemicals in Autoclaved 18.2 mega Ω-cm Water.
Confirmed test for being RNase/DNase and protease free.

PROCEDURE:
Add the reducing buffer to samples: Dilute 1 part sample with 1 part of the reducing or non-reducing sample buffer. More sample buffers can be added if necessary. Dry samples can be dissolved directly into the 2X reducing sample buffer. Then, Heat the sample/sample buffer mixture at 85 á´¼C for 2 minutes for optimal results. Centrifuge for 2 minutes to precipitate undissolved materials and load the samples on the gel.

STORAGE TEMP
Products with Cat. No. BTSSB71 (containing β-mercaptoethanol) are stored at 2-8 ᴼC. the product can be aliquoted into small tubes and stored frozen for extended shelf life.
Products with Cat. No. BTSSB72 (containing DTT, Dithiothreitol) are stored frozen at -20 á´¼C. It is recommended to aliquot the sample buffer in small tubes to avoid multiple freeze-thaw cycles.
