DESCRIPTION
The 6X Laemmli SDS-Sample Buffer without reducing agent is used for preparation of samples for SDS PAGE.
Laemmli SDS sample buffer is used as an electrophoretic dye for denaturation of proteins and monitoring the front of running gel.
Our 6X Laemmli Sample Buffer can be used with both home-made and commercially available precast protein gels such as Mini-PROTEAN® and midi Criterion™ gels from BioRad, Novex Tris-Glycine gels from ThermoFisher, and many others.
The 6X SDS-Sample buffer is composed of 375 mM Tris Base / Tris-HCl, 9% Sodium Dodecyl Sulfate (SDS), 50% Glycerol, and 0.075% Bromophenol Blue (pH 6.8 ± 0.1).
The final concentrations in 1X SDS Sample buffer are: 62.5 mM Tris-HCl, 1.5% Sodium Dodecyl Sulfate (SDS), 8.33% Glycerol, and 0.0125% Bromophenol Blue (pH 6.8 ± 0.1).
Our buffer is formulated from Highly Pure Molecular-Grade Chemicals in Autoclaved 18.2 mega Ω-cm Water.
The sample buffer is evaluated and certified for the absence of RNase/DNase and protease activities.
PROCEDURE:
dilute 1 volume of the 6X sample buffer with 5 volumes of sample solution (Example: add 5 µL of the 6X sample buffer to each 25 µL sample). Reducing agents such as Beta-Mercaptoethanol can be added to the mixture at a final concentration of 1.5%-2.0%, or DTT (Dithiothreitol) at a final concentration of 50 mM.
For dry or precipitated protein samples, dilute the sample buffer to 1X using deionized water and the proper amount of reducing agent. Then dissolve the protein sample in an appropriate amount of the diluted sample buffer.
Samples are heated in gel loading/sample buffer for either 5 minutes at 100 ᴼC, or 10 minutes at 70 ᴼC to aid in the denaturation. At this point, samples can remain at room temperature if they are to be used immediately or placed at 4 ᴼC or –20 ᴼC for later analysis.
